To date, more than 100 imprinted genes have been reported and they are thought to be regulated by the ICR domains (Imprinting Control Region) which are rich in CpG islands and differentially methylated. Methylation of ICR domains within promoters change the structure of chromosomes, resultingin silencing of downstream genes. ICR could also modulate imprinted genes in a long-range control manner.There are two mechanisms of this process described by recent studies.
One of which is the Igf2-H19 example. Igf2 (insulin-like growth factor type2) gene is located 90 kb upstream of the H19 gene which encodes a long noncoding RNA. There is an ICR domain residing 2 kb upstream from the H19 promoter and an enhancer sequence at its downstream. In the paternal gamete, the ICR is methylated leading to the methylation of the H19 promoter in an unclear way. Therefore, the H19 is repressed on the paternal chromosome whereas the Igf2 could be expressed. In the maternal gamete, the ICR domain is unmethylated and bound by the CTCF protein. CTCF is a type of zinc-finger protein which exclusively binds to the unmethylated sequence and acts as an insulator to block the interaction between the enhancer and the Igf2 promoter, resulting in the suppression of the Igf2. Thus, the differentially methylated ICR is associated with the differentially silencing of imprinted genes18.
